Remains of the Day Gonna start writing introduction to TVD paper (copy bits from previous TVD reports) Get figures together for Australia report Prepare Earth Alive figures Since the Oomycete gradient PCR didn't appear to say the problem was temperature (see below), I'm gonna try a nested approach to get amplifiable sequence for oomycete TRFLP in samples where there isn't much. So I used the DC6 primer in combo with OomR plus 5x25 ul of MgCl per 50 ul reaction. 35 cycles and 62 degree annealing. Will take 1 ul of this and apply it to OomF and OomR reaction, run under same conditions for a total of 70 cycles of amplification. Let see on gels tommorow how well this approach works.
