Remains of the Day

  • Repeated PCR of fungal TRFLP from rep 2 of Tilbury TVD tomato rhizospheres (but didn't get chance to run a gel and check)
  • Started strawberry rhizosphere manipulation experiment with Steph. Planted dirty, unwashed speedlings of Albiion in Brian's bad soil (muck soil from carrot field infested with oomycetes), Tilbury 2011 (TVD pathogens I guess), Delhi untreated potato soil (contains Verticillium says George), autoclaved sand, and autoclaved sand pluse a blend of 8 bacteria (5 speedlings were soaked in this blend for 30 minutes, then planted and watered with it). All plants were watered with a miracle grow fertilizer added at a rate of 0.5 g/L, then put in the growth room. The bacteria were grown individually with shaking for two days in 100 mL of R2A broth, then pelleted in a centrifuge for 10 min at 4000 rpm, then resuspended in 0.1 M sodium chloride buffer. The ODs were too high to measure, so we opted just for CFU counts, which were obtained by plating 50 ul of 10,000 and 1,000,000 dilutions of the bugs on R2A media - incubated at 30 degrees. An average of 48 mL of these concentrations of microbes were mixed together to form the multibacteria mix for soaking plants for 30 minutes.
