The Daily Grind

  • Innoculated corn in rhizosphere exiting experiment. Did two reps (two plants) per Arabad::GFP in pDRIVE labelled bacteria, with 1A8, 1C4, 1F7, and 1G6. Innoculum was 5 ul of OD600=0.2 suspension in sodium phosphate buffer, which was pipetted into a vertically cut hole in the stem as it poked out above the tube and parafilm lid. Plants were at the 4 leaf tip stage. As the droplet usually wasn't absorbed into the hole, plants were left lying vertically to dry the droplet off and avoid driping. Plants will be harvested for samples in 5 or 7 days. Plants were grown with in the growth chamber at 22 degrees, with 14 hour days (or whatever the conditions are downstairs).
