The Daily Grind

  • As there was so much contamination in the plates and glycerol stock from the R2A cultured endophytes, I decided to redo the glycerol stocks using sodium phosphate buffer and toothpick swiped colonies instead which were immediately frozen after resuspension. Visually seeing the bacteria being put into the mixture and not amplifying them in media seems like a good idea to reduce contamintation.
  • Using these new "Good" plate 3, I innoculated 500 ul x 96 wells of nitrogen free LGI, LGI plus 10 mM ACC, and R2A plus 10% 1M Glucose. The latter was grown in 37 degrees with shaking, while the two former were grown in 25 degrees with shaking. Will reinnoculate plates with this good glycerol starting tommorow.
