Third observation on cell growth for dilution sequence

Well No.(approx. avg. cell count): Obervations

1(1e6): No significant changes: Continuous dense structure with a reticular structure based on star like extensions of the cells. No distinct colonies. Close to the edge of the well, but not exactly at the edge the cells form a very dense ring enclosing the whole plate.

2(2e5): No significant changes: Continuous less dense structure with a reticular structure based on star like extensions of the cells. No distinct colonies. Same ring structure with high cell density.

3(4e4): No significant changes: Unevenly distributed cells. Regions with close connections between cells and then wider gaps with few individual cells. Agglomerations cannot be described as distinct colonies. More dense ring structure close to the edge, these cells have a lot of connections, followed by an outer ring with only very few cells.

4(8e3): Regions with small cell clusters, also regions with bigger continuos structure and regions with lots of scattered individual cells. Cell clusters have a medium cell density, e.g. <10 cell diameter distance between them. Close colonies tend to grow into each other. Ability to distinct between colonies varies between different regions in the well. More dense outer ring structure close to the edge.

5(1.6e3): No significant changes: Less individual cells than well no. 4. Lots of distinct cell clusters, most with <10 cells. Different degrees of compactness: some are very loose with multiple cell diameter space (<5 cell diameter) between them, some are closer and have connections and some are so close that individual cells are hard to identify.

6(320): No significant changes: Few, unevenly distributed cells. Only few clusters, with loose structure, and a few very compact ones.

7(64): Very few individual cells. Two distinct clusters, very compact, less than 15 cells in one cluster.

8(12): Very few individual cells, no colonies found.

9(~2): No living cell found.

10(<1): No living cell found.
