Summary See protocol here. The past week I transfected BT474, BT549, and MCF10A cell lines with either KAH237 (KAH160 in pMACS LNG.IRES plasmid), or LNGFR control plasmid. I imaged KAH237 cells with phase & mCherry and harvested all cells, along with two negative controls (BT474 & BT549), fixing in 1 mL 4% formaldehyde in PBS. I then measured cell concentration by flow cytometry, rinsed the cells with incubation buffer (BSA in PBS), and stained with 100 µL of CD14-FITC antibody diluted 1:500. Samples are in the 4 °C fridge now.
