ABSTRACT
- Preparation of tubes with DH5α transformed with Pamela Silver Lab's plasmids to make glycerols and plasmid extraction.
- Today I left 3 little test tubes with 3 ml of LB liquid medium, each one with a distinct anbiotic; 3 μl of ampicillin, 3 μl of spectinomycin and 3 μl of chloramphenicol at a concentration of 100 μg/ml, 100 μg/ml and 25 μg/ml, respectively. The tubes were inoculated with the DH5α E. coli that carry Pamela Silver Lab's plasmids. Three additional tubes, with the same amount of antibiotic, were prepared and also put at 37°C and served as negative controls. These tubes are going to be used to make glycerols of the bacteria carrying the plasmids.
- I also prepared 3 test tubes with 5 ml of LB liquid medium, again with the same proportion of their respectives antibiotics, to let incubating the transformed bacteria and let them ready for plasmid extraction.
- All test tubes were left to grow overnight.
