  • Prepared 3 potassium phosphate (KPi) buffers for use in bringing the copper/BSA solution to varying pH points.
1M KPi Buffers, Lower pH

6 6.6mL 43.4mL
7 30.75mL 19.25mL


  • The pH 5.8 buffer was, by dropwise addition of 1N HCl, adjusted down to pH 5. The solutions were not diluted from their 1M concentrations, but were instead used directly...
  • Soln #1: pH 5

- 1.5μM BSA

- 0.25mM cupric acetate monohydrate

- 5mL of 1M KPi at pH 5

  • Soln #2: pH 6

- 1.5μM BSA

- 0.25mM cupric acetate monohydrate

- 5mL of 1M KPi at pH 6

  • Soln #3: pH 7

- 1.5μM BSA

- 0.25mM cupric acetate monohydrate

- 5mL of 1M KPi at pH 7


  • The solutions were placed in glass test tubes and heated at 80°C in an oven overnight.
