Prepare 293T P/S

Pick mOrange Clones

  1. Pick clones 1, 3, 5, 7
  2. Place in 200 mL w Amp for maxi prep
  3. 30C shaker incubator

Make 9-12 sequences

  1. EcoRI digest mCherry and 2,4,6,8 backbones (overnight)

ppGFP plasmids in 384 well for testing

  1. Trypsinize 6 well plate
  2. Count wells
    1. 2
    2. 4
    3. 6
    4. 8
    5. (+) control, fugene
    6. (-) control, cells

Methylate SssI 2, 4, 6, 8
