9/14/2016 Flourescence

Methods

1. Sample was made in a capped cuvette:

  1. pH 5
    1. Myoglobin: 0.25mM Au, 12.5μM Myoglobin, 3mL total
      1. 164μL AuCl3
      2. 30 μL 1M HCl
      3. 274μL Myoglobin
      4. 2532μL Water
  • Myoglobin stock solution was incorrect so the concentration of myoglobin in solution was actually 16.8μM.

2. The cuvette was heated to 80C and samples were taken every 5 minutes using the fluorescence spectrometer for three hours.


3. Samples for future UV-Vis spectroscopy were made using prepared stock solutions with the following concentrations:

  1. AuCl3 :4.57mM
  2. Myoglobin: New stock solution was made
    1. 33.48mg myoglobin
    2. 10.0mL water
    3. MW of myoglobin:16951g/mol
      1. Concentration: 198μM
  3. Fructose: 13.0mM
  4. HCl: 1mM
  5. NaOH: 1mM
  6. NaOH: 1M
