1. We prepared compotent cell (Top 10)

2. We prepared storage for Top 10 (%20 glycerol)

3. We tryed double digestions. The following amounts are added to microtubes;

 *B0010,  
           1) water      20.79 µl
           2) Buffer     3 µl
           3) Enzyme     1;1µl  (EcoR1, Spe1)
           4) DNA        3.21 µl
           5) Fast AP    1µl         =30 µl
 *B0012,   
           1) water      20.95 µl
           2) Buffer     3 µl
           3) Enzyme     1;1 µl   (Xba1,Pst1)
           4) DNA        3.05 µl 
           5) Fast AP    1µl         =30 µl
 *KGF5(11.08.09),  
           1) NEB2       6 µl
           2) BSA        1 µl
           3) Spe1,EcoR1 1;1 µl
           4) DNA        41 µl       =50 µl
 *LUXR-RBS(05.08.09),  
           1) NEB2       6 µl
           2) BSA        1 µl
           3) Xba1,Pst1  1;1 µl
           4) DNA        41 µl       =50 µl
 *BB12(04.08.09),
