13.07.09 1. Storage of transformed cells. Pick single colonies with toothpick and inoculate in 5 ml LB medium (broth) containing appropriate antibiotic. (ampicilin) Amp = 5 µl.Incubator 240 rpm, overnight (16 hours) Centrifuge at 4000 rpm for 10 min. Remove supernatant. Resuspend pellet with 1 ml LB 15% sterile glycerol ( 1ml × %15 = 150 µl) Store at -80 C.
