  • Isolated and cut out from yesterday’s low-melt agarose gel the bands that we believe are VEGF and the yeast plasmid. Ran an amplification reaction on the VEGF band. Backbone band is the isolated product of a digestion reaction, so we placed it in 4 degree storage. After purification of the VEGF, we sequentially digested it using HindIII and SacI. Ran on a gel to test if the reactions worked and to potentially isolate the selected fragment from the digestion for ligation tomorrow.
