  • Found a little bit of growth from the transformation, but Dr. Kozminski was not convinced because growth was not as much as expected. Thus, we acquired five 0.2 micrometer filter sterilizers from Kozminski’s lab, and re-transformed cells with Kozminski’s reagents.
  • Ran three PCA reactions: 8 oligos, 10 oligos, and 12 oligos. Gel analysis showed that the 8 and 10 oligo assemblies worked, but the 12 oligo assembly did not have any results. This could be attributed to the lack of polymerase in the PCA reaction mixture because we were running low on polymerase. Ordered Phusion polymerase.
