July 8th, 2011 Made the master mix again. Performed Gibson assemblies of oligos 6, 7, and 8 at different thermocycler times (15, 20, 30, and 45 minutes). PCR purified one batch, ran a gel electrophoresis. From the top to the bottom: 1. NEB 100bp ladder 2. empty 3-6 were pcr purified: 3. 15 minutes 4. 20 minutes 5. 30 minutes 6. 45 minutes 7-10 were direct gibson products: 7. 15 minutes 8. 20 minutes 9. 30 minutes 10. 45 minutes We saw bands in lanes 7-10 at greater than 500bp and faintly between 100 and 200bp. The color of the DNA was blue under the UV light in contrast to the purple pinkish ladder.
