June 27th, 2011 Viewed the incubated cells and verified that cells only grew on SC plates, which is what we expected because none of the cells should be able to make uracil, leucine, or histidine. Made the master mix and oligo mix. Used the isothermal step to assemble the DNA construct. Purified the DNA from the reaction enzymes in the master mix so they do not interfere with future digestion/ligation procedures. Developed the protocol for thermocycler with Gibson assembly.
