- Mini-prepped cell cultures grown up yesterday. Used Promega standard procedure.
- Made glycerol stocks from cultures made yesterday (Lock 1, Lock 3, and RFP)
- Isolated and inoculated a colony from the GFP plate streaked yesterday. DID NOT use antibiotic. Will make glycerol stock tomorrow.
Mini preps yielded very low concentrations of plasmids, if they had yield at all. Discarded all but the GFP plasmids, because we currently do not have any others. Possible theories for low yield are the presence of antibiotics in the growth media used or the 24 hour gestation period.
