July 13, Monday, 2009 Going to look at pCTCON2-GFP by fluorescence microscopy. Protocol 1. Grow pCTCON2, pCTCON2-GFP transformants to sat'd overnight in sterile-fitted 2x -W / 0.1% glu (so, actual conc. .2%) 2. In the morning, dilute at OD660 to 0.2 - 0.3 in 2x -W/2% gal 3. ~ 3 hours @ 30 degrees C 4. Scope! Restreaked transformants from Thursday, 9 July 2009 b/c plates too dense and pCTCON2 plate accidentally gouged w/ lid. Control plate had mold. Why so much mold? End of the Day
