Transformation of competent cells "Escherichia coli" Today I transform plasmids with iGEM parts in Escherichia coli. Method to transform 1. Unfreeze the competent cells. 2. Put 1 μl of DNA in the eppendorf tube of competen cells and wait 20 minutes. 3. Put the competent cells with DNA in the Termo to 42 °C for one minute. 4. Leave the competent cells five 5 minutes in ice and put 1 ml of liquid LB. 5. Leave the tubes for one hour to 37 °C. 6. Centrifuge the tubes for 30 seconds to 13 mil rpm. 6. Trow supernant and shake the cells in vortex. 7. Take the drop of the tubes and put in the petri dish. 8. Scatter the cells with the bail.
