Competent Cell Test

  • To test the competency of the two batches of DH5a competent cells (Batch ‘A’ and ‘B’)and one batch of BW27783 competent cells
  • One glycerol stock was made from each of the batches by adding:
    • 14uL 100% glycerol
    • 86uL competent cells
    • Freeze in -80 freezer for 30 minutes
  • Each aliquot was transformed following the British Columbia iGEM Transformation protocol using 1uL of 5ng/uL pBADwt+RFP
  • All 3 aliquots were plated on chloramphenicol plates
