Testing Competent Cells In order to test the competence of the DH5α cells, one glycerol stock from each of the four competent cell tubes (A,B,C & D) were made using: 14uL 100% Glycerol 86uL Competent cells Each 100uL aliquot was put into the -80 freezer for 2 hours The 4 aliquots of competent cells were transformed using 1uL of pUC19 following the iGEM:British Columbia Transformation protocol In addition, one aliquot of DB3.1 cells that have been previously tested to be competent were also transformed with the same conditions as a control
