Overview

I took the 55°C and 56°C fractions from the thermal gradient PCR of DBN002 with f2 primer (here) and ran them on a 1% agarose gel, then excised the 1444bp fragment and purified it using the GenElute gel extraction kit.

DNA concentration (n=2):

Sample ID average ng/µL stdev ng/µL
DBN002 16.84 1.261

Yield is low, but should be sufficient for Type IIS ligation.
