New Restriction Digest/Purification of Plasmid and Gel/Extraction of GFP and ATF1
- Today we ran a new digest and purification of the blank plasmid (using the correct kit!!-GeneJet PCR Purification Kit). Also, we used our already digested (with KPN1 and EcoR1/Pst1 enzymes) samples of GFP and ATF1 and combined the samples into one large sample of GFP and one large sample of ATF1. Next, we set up a 1.2% gel ( two wells were taped together to make one big well for the GFP sample and the same was done for the ATF1 sample) to run the samples. After cutting both bands out of the gel we performed a purification of the DNA using the GeneJet Extraction Kit. For the Tuesday after Thanksgiving break (11/29/11) the purified and digested samples of GFP, ATF1, and the plasmid will be ligated together.
