GelGreen Gel Electrophoresis

I am performing an agar gel electrophoresis to try and visualise a DNA ladder. I am using an iorodeo gel box. Gel was prepared as follows:

  • 4g Agarose
  • 200mL 1 x NaOH/Borax buffer

Heated in microwave until melted. Added 20ul Gelgreen when solution had cooled slightly. Mixed thoroughly by agitation and with a pippette tip. Cast gel. Filled chambers with 1 x NaOH/Borax buffer. Ran at 60mA and 100V for 1 hour.

It worked, but after a while it wasn't separating well because the ladder was large and the gel was 2%. Visualisation was quite good with the filter on top.
