Gold Nanoparticles wrapped in protein

  • This experimentation is in continuation from the previous year and is based off of the investigation of Bakshi, et al. Bovine serum albumin (BSA) was found to form nanoparticles with gold (Au). BSA acts as a reducing agent by converting Au (III) into Au (0). When heated, BSA unfolds and the amino acid, cysteine, reacts with the hydrogen bonds of chloroauric acid, HAuCl4. AuNPs in BSA were synthesized at a variety of mole ratios and serve as the platform for the rest of the reactions and experiments conducted
  • Various methods of synthesizing AuNPs have been studied from BSA, HRP, Lysozyme, and Adenosine Deaminase. Various methods have also been employed to measure the amount of or concentration of gold nano-particles present including UV-Vis and Atomic Absorption.
  • Three activity assays were conducted. The first two were to look at the activity of Horse Radish Peroxidase (HRP). One was HRP with aminoantipyrine and the second was HRP and luminol. The third activity assay was an assay conducted to test the activity of adenosine deaminase (ADA).
  • Adenosine Deaminase was synthesized in lab from E.Coli cells, purified using FPLC, and the concentration of the protein extracted was determined through the conduction of a Bradford Assay.

  • This round of experimentation began in August of 2012 and continued until the end of November 2012.
