Objective

We are running the same procedure as yesterday only with an inhibitor, EHNA.

Description

The absorbance graph is saved every 15 seconds for a period of 10 minutes total. Using the following equations, concentrations of Adenosine and Inosine with the inhibitor are calculated.
Absorbance at 249nm=Molar absortivity of Adenosine at 249nm * concentration of Adenosine + molar absortivity of Inosine at 249 * concentration of Inosine
Absorbance at 259nm=Molar absortivity of Adenosine at 259nm * concentration of Adenosine + molar absortivity of Inosine at 259nm * concentration of Inosine
Molar absortivities are calculated from the the first lab in september's data.
Molar absortivity of Inosine at 249 nm is 1107 M-1cm-1.
Molar absortivity of Inosine at 259 nm is 6380 M-1cm-1.
Molar absortivity of Adenosine at 249 nm is 14500 M-1cm-1.
Molar absortivity of Inosine at 249 nm is 13280 M-1cm-1.

Data

EHNA stock

  1. 5mg EHNA in 1mL DMSO ---> 15.9mM EHNA
  2. (1.9μL)(15.9mM EHNA)=(10mL)(C2). C2 ---> 3μM EHNA

The reaction samples will contain roughly 1nM EHNA
