Objective

  • Filter hydrogels and observe under microscope
  • Observe indicator color changes with change in pH

Procedures

Part 1: Filtering hydrogels

1) Pour top layer of safflower oil out of vials A1, B1, B2, C1, C2, D1, and D2

2) Add hexanes to each vial and vortex

3) Filter each hydrogel sample using vacuum filtration

4) Combine samples B1 and B2, C1 and C2, and D1 and D2

5) Re-disperse filtered hydrogels in phosphate buffer

Part 2: Observation of the addition of an acid/base to hydrogels containing indicators

Notes

  • For next time. Sonicate hydrogels containing dye using super sonicator. Filter the (hopefully) milky solution first through regular filter paper. Then, taket he filtrant (whatever is in the flask) and filter through nylon filter paper.
