Objective

  • To create standard solution of gold for calibration of the atomic absorption spectrometer. The students will measure the gold content of their BSA nucleated AuNPs against these standards.

Bench Work

  • 1000 ± 10μg of Au/mL of solution ( 10 % v/v HCl) for the stock solution
    • from Inorganic Ventures cat# AAAU1-1, lot # F2-AU04012
    • the solution is certified by ICP (ion couple plasma spectroscopy) to NIST SRM 3121

For the AAS (atomic absorption Spectrometer), the published detection limit for gold is 0.2 ppm ( a 1% absorbance) for this instrument. recent experiments put the detection limit closer to 2 ppm for gold, and since 20 ppm is roughly 0.1 mM Au, therefore I created standards ranging from 5 to 40 ppm.

  • created 50 mL of each standard: 5, 8, 10, 15, 20, 25, 30, 40 ppm Au
    • from the 1000 ppm Au stock standard
    • using 50 mL volumetric flasks
    • all diluted in 1 M HCl
