Procedure
Yeast PCR Lyticase lysis bufferLyticase buffer(Kwiatkowski et al, 1990)
To 5 mL total volume with millipore water (4.67 mL). Aliquot and store at -20 °C. ProcedurePick a small amount of each colony into 15 uL lysis buffer, suspend. Heat 60 min at 37 °C then 10 min 95 °C.
