*According to the M9 medium preparation protocol, mix:

**glycerol and
**M9 salts.
**Dip the pH-meter probe in the medium to monitor pH (wash probe with deionized water and calibrate pH-meter with 2 points when required);
** ajust pH with HCl, NaOH (or KOH). While handling strong acid/base, always wear suitable protections!
** wash the pH probe with deionized water and store it in KCl 3M.
* Autoclave the mix in a bottle.
* Equilibrate at room temperature and add the remaining ingredients (casamino acids, thiamine, CaCl2, MgSO4).

pH adjustment for LB medium

  • Before autoclaving the LB medium:
    • dip the pH-meter probe in the medium to monitor pH (wash probe with deionized water and calibrate pH-meter with 2 points when required);
    • ajust pH with HCl, NaOH (or KOH). While handling strong acid/base, always wear suitable protections!
    • wash the pH probe with deionized water and store it in KCl 3M.
  • Autoclave the medium

  • According to the M9 medium preparation protocol, mix:
    • water,
    • glycerol and
    • M9 salts.
    • Dip the pH-meter probe in the medium to monitor pH (wash probe with deionized water and calibrate pH-meter with 2 points when required);
    • ajust pH with HCl, NaOH (or KOH). While handling strong acid/base, always wear suitable protections!
    • wash the pH probe with deionized water and store it in KCl 3M.
  • Autoclave the mix in a bottle.
  • Equilibrate at room temperature and add the remaining ingredients (casamino acids, thiamine, CaCl2, MgSO4).
