Procedure

  • Add all 100 or 150uL of an MC1061 competent cell tube to 400mL LB
  • Grow up overnight on 37*C shaker
  • split into two centrifuge bottles and centrifuged at 4200rpm for 14 minutes
  • pour off supernatant
  • wash each pellet with 10mL artificial sea water (ASW)
  • pour off ASW
  • add 10mL ASW to each bottle and resuspend pellets
  • centrifuge at 4200rpm for 14 minutes
  • pour off supernatant
  • re-suspend pellets
  • combine the contents of the two bottles
  • create 2uL aliquots
