Entry titleOn tuesday of this week we began to digest and purify our plasmids we have extracted to prepare them for ligation. we also have bacterial cultures growing but we have yet to see any real growth from any of them, we will allow them to continue to grow until tuesday (october 30th) in hopes that they will show signs of growth by then. Thursday we continued with the purification of the plasmids using the PCR purification kit. we are running a gel to see what we get as well. also on Thursday we made a 4th culture of our bacteria because of the lack of growth in the original three, this one will be on a rotation plate while it is left over the weekend to see if that will make a difference.
