Summary

A repeat of the previous experiment.

Materials & Methods

Materials

  • petridishes (10 mL)
  • 6-wells plates (2 mL)
  • 24-wells plates (0,5 mL)
  • DMEM S+

Method

  • Cells are collected in DMEM S+ as described in Splitting cells and counted as described in Counting cells
  • Cells are split over several plates
    • 4 petridishes (10 mL) receive 500.000 cells
    • 1 6-wells plate (2 mL) receives 100.000 cells per well
  • 2 24-wells plates (0.5 mL) receive 20.000 cells per mL

Results

Cultures contained 57.25×104 cells per mL (17175×103 total)

  • 6-well (2 mL per well)
    • 12.78 mL DMEM S+
    • 1.22 mL cell suspension
  • Petridish (10 mL per plate)
    • 45.64 mL DMEM S+
    • 4.36 mL cell suspension
  • 24-well (0.5 mL per well)
    • 24,3 mL DMEM S+
    • 1.7 mL cell suspension
