Objective

  • Create Tris and phosphate buffers
  • Titrate buffers using pH and conductivity meter
  • Redo preparation of AuNP with lysozyme since previous stocks did not produce fibers

Description

The procedure for the creation of buffers can be found here.
\frac{121.14g}{mol}|\frac{.01mol}{1L}|\frac{.500L}{1}=0.6057g of Tris was used to create a 500mL stock of Tris buffer.

For the creation of the phosphate buffer, this link was used to determine the amount of sodium phosphate (dibasic) and sodium phosphate (monobasic) needed for a 500mL stock.

The stock solutions of AuNP with lysozyme were prepared using this protocol.
