Steve Koch 15:07, 18 April 2009 (EDT): I found a presentation from 2001 showing that I'd tried out sucrose unzipping. I uploaded this to Google Docs: http://docs.google.com/Presentation?id=dgqjkh6p_46wckm4f3

Preliminary data showed for (among others):

  • Unzipping BsoBI in sucrose (osmolite)
  • Unzipping cross-linked protein-DNA (UV cross-linking of BsoBI and XhoI)
  • Overstretch fluctuations
  • Unzipping BsoBI in high calcium (shows increased unzipping force, and increased non-specific binding)

I should start uploading all of them, but I don't want to get too distracted now.
