Objective

  • Continue SDS-PAGE analysis from 10/25.
  • Perform gold nanoparticle(AuNP) synthesis to produce purple fibers, as well as determine if protein fibers form in the absence of gold.

Description

  • Destain Gel

1. Remove Coomassie Blue stain

2. Add destaining solution

3. Leave on shaker


  • AuNP Synthesis: 15 μM Bovine Serum Albumin (BSA), 0.75 μM Chloroauric Acid (HAuCl4), and H2O

1. Tube 1: 1 mL BSA (15 μM) , 2 mL HAuCl4 (0.75 mM), 7 mL H2O

2. Tube 2: 1 mL BSA (15 μM) , 9 mL H2O

3. Tubes placed in oven set at 80 degrees Celsius, and removed for 10 minutes every 30 minutes over the course of 2 hours.

Data

  • AuNP Synthesis:

Tube 1 was a clear solution, with a gray and green protein fiber aggregate.

Tube 2 remained clear.


  • SDS-PAGE:

Results indicate that while the protein of interest was isolated, it was not pure enough for further experimentation.

Notes

  • Molar ratio of Au/BSA for Tube 1 - 100.
  • Tube 2 contained no gold.
  • Further experimentation is needed to understand what might have happened in these experiments.
