ObjectiveRun our samples using ICP, measure conductivity for each concentration and Collect Uv-vs data for the different concetraions. DescriptionFifteen supernatant solutions of 20-mL-fiber sample are removed; so 3 x (blank, 1 µM, 100 nM,10 nM, and 1 nM sample). Four of the supernatants are saved in separate falcon tubes for conductivity and ICP measurements. The 3 blank jars are only filled with a certain volume of tris while the rest 12 jars are filled with the appropriate amount of tris and protease. Blanks and sample are heated and placed in falcon tubes. The 15 tubes of blank and sample along with the 4 supernatant tubes are centrifuged for 5 minutes. The ICP standards are used to measure their conductivity. The conductivity meter is calibrated with different pHs such 9 , 7 and 4. After calibration, a clean conductivity meter stick is placed in each solution to record conductivity measurements. The stick is rinsed with distilled water and dried using kimwipes between after each measurement. Data AnalysisObservationsthe ICP data shows that the higher the concentration of protease present in the solution the higher the gold concentration For conductivity, we see that the cloudy samples (a blank and a10-nM) have the same conductivity . while others are showing some kind of trend so increasing the protease concentration would increase the conductivity of the gold.
