Love light, do it again! Today, I arrived early to grow some colonies of the click beetle luciferases, which I check and obtained one that was functional. I am planning on following 2 different ligations to have luciferase and LRE in the same plasmid. The first strategy involves ligating the luciferase and its constitutive promoter into the plasmid containing the LRE; whereas the second one involves ligating LRE into the plasmid containing the luciferase.
