## Objective

• took florescence of the differ concentrations used with hydrogel pieces and determined the concentrations/mols of dye attached

## Description

NOTE: Mass of whole hydrogel: 12.1419 grams, piece of hydrogel 0.8 grams, so instead of 0.193 grams to occupy all the butayne-1-ol sites(0.000017mols) an amount of 0.014 grams of dye is needed.

• the stock solution of dye is 5mg/ml=5g/L therefore the amount of grams of dye in vial, as well as assuming 1:1 reaction between butane-1-ol and dye then a successful reaction will have a percent reacted ~ equal to the percentages below. Percentages below are equal to the amount of dye in mols added to the film/the amount of mols that should react with the butane-1-ol times 100.
• 1µL of Dye= 0.000005g= 8.9E-9 mol/0.000017mols= 0.0523%
• 10µL of Dye= 0.00005g=8.9E-8 mols/0.000017= 0.523%
• 100µL of Dye= 0.0005g=8.9E-7mols/.000017= 5.23%
• 200µl of dye= 0.001g= 1.8E-6mols/.000017= 10.5%
• 500µl of dye= .0025g= 4.4E-6 mols/.000017= 25.8%

• after soaking for 24hours with gel: 1.34E-9 mols
• leached in water solution after 24 hours: 4.3919E-9 mols

• 7.56E-9mols - 4.3919E-9mols= 3.17E-9 mols of dye in hydrogel actual
• mols butyne-1-ol sites in small piece of hydrogel (assuming 1:1 reaction): 0.000017

• Percent of dye reacted with butyne-1-ol= 6.94%

• starting solution: 4.4E-6mols
• after soaking for 24hours with gel: 8.8E-7 mols
• leached in water solution after 24 hours: 8.4E-7 mols
• 4.4E-6 mols- 8.8E-7mols= 3.5E-6 mols of dye soaked up by gel in 24 hours
• 3.5E-6mols - 8.4E-7mols= 2.67E-6 mols of dye in hydrogel actual
• mols of butyne-1-ol sites in small piece of hydrogel: 0.000017
• Percent of dye reacted with butyne-1-ol= 15.71%
