Stock Solution Preparation

1. Prepare 100 mL 2.5 mM HAuCl4 at pH 3.6

  • 2.5 mM HAuCl4 * 14307 g HAuCl4 / mol * 100 mL = 0.085 g HAuCl4
    • 0.08533 g was the final amount weighed giving a final concentration of 251 μM HAuCl4
  • Add 0.08533 g HAuCl4 to a volumetric flask and dilute to 100 mL with deionized H2O
    • Monika prepared a total of 100 mLs HAuCl4 for both groups.

2. Prepare 50 mL 20 μM Lysozyme

  • 20 μM Lys * 340 g Lys /mol * 50 mL = 0.014 g Lysozyme
    • 0.01417 g was the final amount weighed giving a final concentration of 19.81 μM Lysozyme
  • Add 0.014 g Lys to a volumetric flask and dilute to 50 mL with deionized H2O

Gold Colloid and Fiber Preparation

1. 30:1 Au:Lys Colloid

  • To prepare 5 mL add 0.5 mL 251 μM HAuCl4
  • Desired Lysozyme concentration= 250 μM HAuCl4 / 30 = 8.3 μM Lys
    • M1V1=M2V2
    • 19.81 μM * V1 = 8.3 μM * 5 mL = 2.095 mL Lys
  • Dilute with 2.405 mL deionized H2O
    • Final volume = 5 mL
  • Heat at 80°C for 4 hours

2. 45:1 Au:Lys Fiber Formation

  • To prepare 5 mL add 0.5 mL 251 μM HAuCl4
  • Desired Lysozyme concentration= 251 μM HAuCl4 / 45 = 5.56 μM Lys
    • M1V1=M2V2
    • 19.81 μM * V1 = 5.56 μM * 5 mL = 1.40 mL Lys
  • Dilute with 3.10 mL deionized H2O (final volume = 5 mL)
  • Heat at 80°C for 4 hours

There were a total of 10 30:1 Au:Lys colloid and 10 45:1 Au: Lys fiber test tubes prepared.
