WetLab Extracting DNA from 2010 distribution. Plate 1. 7A. Plasmid pSB1T3 with 10μL H2O millinQ Transform DH5α competent cells. 1.-Cells defrosted on ice. 2.-100μL of cells for 3μL of DNA from plate. 3.-3 tubes were generated and transformed. 1 control with 50μL of cells. 4.-20 minutes on ice. 5.-Thermal shock at 42°C, 1 minute. 6.-5 minutes on ice. 7.-Add 1 ml of LB at each tube. 8.-1 hour at 37°C. 9.-Plate on Tet30-LB medium Odd cells stored at 4°C.
