Check Transformation I check the plate with the transformed cells with yesterdays ligation between the digested PCR products pSense66(2.3kb) and pBR322(790kb). All sites are PstI digested and purified. I had no growth on the plate. PCR Gel result of 2.3kb product I ran out yesterdays PCR product on a 1% gel to see if I had any new result using the same conditions as in the beginning of the week. In addition I also ran out the ligation samples and yesterdays miniprep. Lane 1: 10µl 1kb NEB Quick Ladder Lane 2: 50µl Pcr product, expected 2.3kb (template pUA66katE) Lane 3: 50µl Pcr product, expected 2.3kb (template pUA66katE) Lane 4: 5µl pSense66 (pcr product digested and purified from earlier in the week) Lane 5: 5µl pBR322 (pcr product digested and purified from earlier in the week) Lane 6: 5µl pBR322 (pcr product digested and purified from earlier in the week) Lane 7: 5µl pUA66katE (gel check of yesterdays miniprep) Lane 8: 5µl pUA66katE (gel check of yesterdays miniprep) Error: Again I have no result from the PCR reaction!!!
