Remains of the Day Gels of phylosphere and gradient PCR suggest temperature is not why I previously had clean products, it was more likely because I only did 35 cycles before, not 40. Therefore, I will use the old phylosphere amplicons (the 35 cycle ones) and try my luck at TRFLP. The PCR Im doing now has 52 degree annealing, 40 cycles and 1 ul of DNA per 25 ul reaction.
