The Daily Grind

  • Cut up potatoes and started 1O1 and 2O2 (empty BAC expressing GUS) O/Ns. Used 100 mg/mL of chloramphenicol for the 4 mL LB tubes, and innoculated with 100 ul of 0.1 OD600 suspensions in LB. That was done at 7 pm and the cultures are shaking now at 37 degrees overnight (for 24 hours).
  • Plated ligations of GFPuv (amplified from pDSK-GFPuv) ligated to either the weak or medium promoters in pSB1A2. Will put in ACC, alsSD, and the RNAi constructs afterwards.
