WetLab: LovTap's promoters Ligation LovTap + promoters LovTap (colony_1) was ligated to seven promoters with different strenght: J23_102, J23_105, J23_106, J23_110, J23_114, J23_116, J23_117. The promoters are in the the plasmid J61002[Amp], these were digested with SpeI and PstI. and desphosphorylated. LovTap was digested with XbaI-PstI Ligation Protocol DNA(LovTap) ..............6μL Plasmid (promoter) .....2μL Buffer 10X T4 Ligase ...2μL Enzyme T4 Ligase .....1μL HPLC ........................9μL TOTAL 20μL The mixture were left ON @ 16ºC
