The α4β1 integrin can mediate leukocyte adhesion to casein and denatured protein substrates

Notes

  • Shows that α4β1 integrin can interact with casein and denatured albumin.
  • α4β1 has a binding tripeptide, Leu-Asp-Val (LDV).
  • Bovine α-casein does not have a LDV site. But, it does have an aspartic residue that is next to hydrophobic amino acids.
    • This is the take home from the paper.
  • They note that you need divalent cations for stuff to stick.
  • They also note that because you need divalent cations for stuff to stick, this is a big indicator that an integrin is involved in the adhesion.
    • I'm not clear on this.
  • The possibility of using α4β1 as casein blocker for motility experiments is pretty high. I should look into it.
  • They finally note that they used two types of casein fragments, one at 6 kDa and the other at 16 kDa. There are no 16 kDa casein components and so I think that they were looking at some other component in this study.
  • The paper was all about α4β1 and how they should look at it more closely to figure out what other kinds of things this integrin will stick to.
