Experiment

Objective Injections on ant vs posterior facial and cochleovestibular.

Procedure

  • Right side: Anterior VII (DiI) vs Posterior VII (DiO)
  • Left side: Cochlear branch of VII (DiI); other posterior component (DiO)

Notes May have a bit of die on VIII - there seems to be a spot of DiI spill at the entry of IX. Injections otherwise looked really clean, especially the cochlear portion of VIII was nicelly lableled in red with no apparent crossing to other haldf. The other half nicely labelled with DiO. On the facial, injections also nice - nice labelling with DiI nicely localised - not so sure about DiO.

Results

Facial - posterior branch

Dont seem to have afferent labelling -

Both columns of motoneurons are labellled.

Facial Anterior branch

(red)

  • Afferents

More clearly labelled - a few fibres, somewhat defasciculated. The rostral bundle is quite short, probably extending to the r3/r2 border, and not too fasciculated. There is a little branch going medially that might be the axons of motoneurons going in to meet the nerve entry. If this is the case, then those axons are coming from r3. If they are afferents, they are going a funny route. The caudal branch is clearly more defasciculated and extends until the entry of IX - it is also in a more medial position that the rostral one.

Beautiful labelling throughout the 2 motoneuron columns on both rhombomeres


  • Other

there seems to be an interesting meshwork of axons crossing coming and going at the level of the entry of the nerve. I also wonder if the same BMN are projecting through both branches - the red and green label do not overlap on the combined image, but I have a bit of a gut feeling that they might be a bit displaced from each other. Need to check this a bit more carefully.

VIII - posterior vestibular

  • Afferents

Nicely labelled - caudal fascicle appears a bit more medially positioned than rostral fascile. Be careful, there might have been some spill into VIII ganglion

No labelled efferents

  • Other

VIII posterior cochlear

(red) (see notes - contamination with IX)

Two clear bundles, with caudal fascicle in a more medial position

  • Efferents

Can defnitely see efferents, but few and far between - seem to be on both r4 and r5, although probably more in r5. Not that many of them!

  • Other

There doesnt seem to be segregation in the afferent path of the 2 branches, although would need to look at this more carefully in the stacks.
