1. Four flasks of LB Broth was prepared.

Lard 1, TLiA, Lard 1+ABC and TLiA + ABC were put into flasks one by one. Also appropriate antibiotics were add into flasks.

We placed these flasks into shaker at 37 °C and 160 rpm.

2. Plasmid Dna isolation and measuring the plasmid Dna content with the Nanodrop.

3. Preparing the ligation reaction.

4. After ligation transformation to the E.coli has done.

The results are positive but efficiency is not as we expect.
