18.07.20091. Measurement of OD with spectrophotometer (600 nm) Dilution factor : 1/5 OD results; Backbone 1 = 0.098 Backbone 2 = 0.038 Backbone 3 = 0.034 They were very low, so we will make plasmid isolation again. 2. Preparation of Electrophoresis Gel 0.5 gr Agarose + 50 ml 1×TAE buffer + 4 µl EtBr We added EtBr after cool down. TAE electrophoresis buffer = 50× We used 1x TAE. (20ml TAE + 980 ml dH2O) 3. Electrophorese was done. Ladder = 5 µl. 2 µl plasmid + 3 µl dH2O + 1 µl loading dye |
