9.8.10 Digestions of each of the minipreps from 9/4/10 was conducted. Each reaction was 50 uL and included digestions of , pgolS + E0420, pgolS + E0430, golB + LamB, and gesABC. This was done as a test to see if the inserts were in the plasmid. A electrophoresis gel was then ran to analyze the digestions. Some of the uncut plasmids were ran and also some gels ran without enzyme, or gels without template were ran as controls to the gel results. A PCR was then done of gesABC and golT using VF2/VR was done because they are the only two inserts that showed results. In the end from this veryification test using VF2/VR only bands were observed in lanes 5-8 which was golT.
